Effects of 2,2¡¯-Azobis (2-amidinopropane) hydrochloride on Plasma Antioxidants in Rat

Abstract
The effect of 2,2¡¯-azobis (2-amidinopropane) hydrochloride (AAPH), a peroxyl radical initiator, on the level of plasma antioxidants was examined in rats. Intraperitoneal administration of AAPH (50 §·/§¸) to rats has resulted in the depletion of total thiols and uric acid (P<0.05) form plasma. The declined thiols level was replenished soon, however, the uric acid level was not restored until 24 hrs after the AAPH administration. On the contrary, ascorbic acid and bilirubin contents were not virtually changed. AAPH treatment led to oxidation of proteins as well as lipids that exist in blood plasma and has resulted in the increase of thiobarbituric acid (TBA) reactive substances. Such changes in their contents were enhanced in a dose dependent manner. Prior administration of ascorbic acid (before 1 hr of the AAPH injection) has effectively suppressed the above oxidative damages, however, notable protection against AAPH intoxication was observed only with large dose of ascorbic acid administration (500 §·/§¸). The changes in the level of plasma antioxidants during the initial stage of peroxyl radical attack show that the first line of defense may be provided by thiols and uric acid. These results suggest that this system my be utilized as in vivo assay method for the biological activities of antioxidants.
